双参通冠方对心肌梗死模型大鼠VEGF表达和炎性因子的影响

摘    要:目的 研究双参通冠方对急性心肌梗死模型大鼠的VEGF表达和炎性因子的影响.方法 SD大鼠分为模型组、空白组、双参通冠低剂量组(2.5 g/kg)、中剂量组(5 g/kg)、高剂量组(7.5 g/kg),采用结扎左冠状动脉前降支建立大鼠心肌梗死模型,灌胃给药2周后,取大鼠的心肌组织,Western blot法检测大鼠的心肌组织VEGF蛋白表达,Elisa法检测心肌组织匀浆中IL-1、IL-6、TNF-α、CRP的含量.结果 与空白组比较,模型组大鼠心肌组织中VEGF表达增高,心肌组织匀浆中IL-1、IL-6、TNF-α、CRP的含量增高,差异有统计学意义(P<0.05);与模型组比较,双参通冠方低剂量组、中剂量组和高剂量组VEGF表达增多,双参通冠方低剂量组IL-6、TNF-α含量均降低,双参通冠方中剂量和高剂量组IL-1、IL-6、TNF-α、CRP含量均降低,差异有统计学意义(P<0.05);与双参通冠方中剂量组比较,低剂量组IL-1、IL-6、CRP含量增高和VEGF表达降低,高剂量组TNF-α、CRP含量降低和VEGF表达增高,差异有统计学意义(P<0.05).结论 双参通冠方可减轻大鼠心肌炎性反应,促进血管内皮细胞增殖,诱导缺血部位血管新生,对心肌具有保护作用,可能与增加VEGF的表达和抑制心肌的炎性因子有关.

Abstract:Objective To study the effects of Shuangshen Tongguan decoction on inflammatory factor and VEGF expression in rats model with myocardial infarction. Methods SD rats were divided into model group, blank group, Shuangshen Tongguan decoction low dose group (2.5 g/kg), middle dose group (5 g/kg) and high dose group (7.5 g/kg). Myocardial infarction model was established by ligation of left anterior descending coronary artery. After two weeks of intragastric administration, the myocardial tissue of rats was taken. Western blot method was used to detect the expression of VEGF protein in myocardial tissue of rats, and the content of IL-1, IL-6, TNF-α, CRP were detected by Elisa. Results Compared with the blank group, the VEGF expression of model group in rat myocardial tissue was increased, IL-6, IL-1, TNF -α and CRP content in the myocardial tissue homogenate was increased, the difference was statistically significant (P<0.05); Compared with model group, the VEGF expression of Shuangshen Tongguan decoction low dose group, middle dose group and high dose group was increased, IL-6 and TNF-α content of Shuangshen Tongguan decoction low dose group was decreased, IL-1, IL-6, TNF-α and CRP levels of Shuangshen Tongguan decoction middle and high dose group were decreased, the difference was statistically significant (P<0.05);Compared with Shuangshen Tongguan decoction middle dose group, IL-1, IL-6 and CRP level of slow dose group was increased and the VEGF expression was reduced, TNF-α and CRP of the high dose group were decreased while the VEGF expression was increased, the difference was statistically significant (P<0.05). Conclusion Shuangshen Tongguan decoction could reduce myocardial inflammatory reaction of rats, promote vascular endothelial cell proliferation, induce ischemia parts of angiogenesis and protect myocardial. This may be related to increased expression of VEGF and inhibition of inflammatory factor in myocardium.
