林蛙皮的酶解工艺及抗氧化作用

摘    要:
目的 优选林蛙皮酶解制备工艺,并研究其抗氧化作用.方法 采用酶解法,通过单因素考察试验和L9(34)正交实验,以水解度作为考察指标,对酶解工艺进行筛选;通过林蛙皮酶解液对羟基自由基,超氧阴离子及DPPH自由基的清除作用,研究其抗氧化能力.结果 林蛙皮的最佳酶解工艺为:料液比1:15,pH 6.5,中性蛋白酶加入量为3000 U/g,45℃下水浴2 h,90℃灭酶10 min,离心,得林蛙皮酶解液,此酶解液对羟基、超氧阴离子及DPPH自由基的清除率在一定范围内随浓度的升高而加强.结论 通过正交实验,得到中性蛋白酶酶解林蛙皮的最佳生产工艺,工艺合理可行,其酶解液具有良好的抗氧化活性,为林蛙皮进一步的化学及药理研究奠定了基础.

Abstract:
Objective To optimize the enzymatic preparation process of Rana skin and study its anti-oxidize effect. Methods Take enzymatic method, use single factor experiment and L9 (34) orthogonal experiment. Evaluating indicator is the degree of hydrolysis to screen enzymatic hydrolysis process; the scavenging activities of hydroxyl radical, superoxide anion and DPPH radical were studied by enzymatic hydrolyzate of Rana chensinensis. Results The optimal conditions of enzymatic hydrolysis process of rana skin were as follows: solid-liquid ratio 1:15, Ph6.5, neutral protease dosage of 3000U /g, at 45 ℃ water bath for 2 hours, 90 ℃ enzyme inactivation 10min, centrifugation and Rana skin hydrolyzate can be produced.The clearance of hydrolysis is enhanced with the the concentration of hydroxyl, superoxide anion and DPPH free radicals in a certain range. Conclusion The optimum production process of neutral protease hydrolysis of Rana chensinensis was obtained through the orthogonal experiment. The process was reasonable and feasible. The hydrolyzate has good antioxidant activity and lays the foundation for further chemical and pharmacological studies.
