赤芍与白芍抗血小板凝集作用的UPLC-MS代谢组学初步研究

摘    要:
目的:利用代谢组学方法研究赤芍和白芍的抗血小板凝集功能及对大鼠内源性代谢物的影响.方法:赤芍和白芍各给药6d后,观察赤芍和白芍对大鼠血小板凝聚功能的影响,并采用超高效液相色谱-电喷雾质谱(UPLC-MS)技术分析大鼠血清代谢物指纹图谱,采用偏最小二乘判别分析法研究给药组与正常组之间的代谢物组差异,寻找可能的生物标识物.结果:连续给予赤芍和白芍提取物6d后,能显著降低大鼠血小板最大凝集率水平,且赤芍组与白芍组组间有明显差别(P<0.01).α-酮戊二酸、苹果酸、白细胞三烯A4、前列腺素E2、前列腺素F2.等内源性生物标识物对于表征赤芍、白芍的抗血小板凝聚作用具有重要的作用.结论:代谢组学研究可以深人探讨赤芍和白芍的抗血小板凝聚机制.
