骨桥蛋白与ras基因蛋白在喉鳞癌中表达意义及相关性

摘    要:目的探讨骨桥蛋白(osteopontin,OPN)与ras基因蛋白在喉鳞癌组织中的表达意义及二者相关性。方法采用免疫组织化学染色方法检测42例喉癌组织(喉癌组)与25例癌旁组织或声带息肉组织(对照组)中OPN与ras基因蛋白的表达情况,分析其与临床病理学特征的关系。结果喉癌组OPN和ras基因蛋白阳性表达率分别为54.8%和73.8%,明显高于对照组(24.0%和48.0%)(P〈O.05);喉癌组OPN阳性表达率在肿瘤T分期、分化程度及淋巴结转移上差异有统计学意义(P〈0.05),在患者年龄、临床分期上差异无统计学意义(P〉0.05);ras基因蛋白阳性表达率在肿瘤临床分期、淋巴结转移上差异有统计学意义(P〈0.05),在患者年龄、T分期、分化程度上差异无统计学意义(P〉0.05);OPN阳性表达与ras基因阳性表达呈正相关(r=0.329,P=0.033)。结论OPN和ras基因蛋白在喉癌的发生、发展中起重要作用,可能成为喉癌生物学特性及评价预后的重要标记物。

Abstract:Objective To study the significance of the expressions of osteopontin (OPN) and ras in laryngeal squamous cell carcinoma (LSCC) and their correlation. Methods The expressions of OPN and ras were detected by using immunohistochemical staining method in 42 cases of LSCC (LSCC group) and 25 cases of adjacent tissue or vocal cord polyps (control group). The relationship of OPN and ras protein expression with the major clinicopathological changes were analyzed. Results The positive expression rates of OPN and ras protein were 54. 8% and 73.8% respectively in LSCC group, which were significantly higher than those in control group (24.0% and 48.0%) (P〈0.05). There was a significant difference in the positive expression rate of OPN protein in T stage, differentiation grades and lymph node metastasis of LSCC (P〈0.05), and no significant difference in patient's age and clinical stages (P~0.05). There was a significant difference in positive expression of ras protein in the clinical classification and lymph node metastasis of LSCC (P〈0.05), and no significant difference in the patient's age, T stage and cell differentiation degree (P〉0.05). The positive expression of OPN was positively correlated with the positive expression of ras (r = 0. 329, P = 0. 033). Conclusion The expressions of OPN and ras play important roles in tumorigenesis and development of LSCC, and could be used as important molecular markers in evaluating the prognosis of LSCC.
