摘    要:目的为改善染料木素的治疗效果,对其进行结构修饰。方法通过成酯、硝基化反应,在染料木素的7-位,4’-位进行修饰,得到一个硝基酯类化合物;并考察得到的化合物对MC3T3-E1细胞的促增殖作用。结果得到的化合物经核磁、质谱等结构鉴定与设计目标一致。该化合物能够显著促进MC3T3-E1细胞的增殖,且呈一定的时间、剂量依赖关系。结论染料木素7,4’-硝氧基丁酸酯较母体药物更好地促进成骨细胞的增殖,为骨质疏松症治疗药物的开发提供新的参考。

Abstract:Objective To improve the efficacy of genistein by modifying its structure. Methods The 7,4’-OH of genistein was modified by esterification and nitration to obtain a nitryl ester compound. The effect of the new product on MC3T3-E1 cells was also observed. Results The obtained compound was exactly the one we had designed. It significantly promoted cell prolification in a time- and dose-dependent manner. Conclusion Genistein (7,4’- nitrooxybutyrate) shows better effect on cell proliferation than its parent drugs. The synthesis of genistein (7,4’- nitrooxybutyrate) may afford a new method in developing antiosteoporosis drugs.
