枸杞多糖联合氟尿嘧啶对荷瘤小鼠免疫功能影响的实验研究

摘    要:目的探讨枸杞多糖单独应用及与氟尿嘧啶联合应用于荷S180小鼠的抗肿瘤机制。方法小鼠右前侧腋窝接种S180肿瘤细胞,制造荷瘤小鼠模型。实验分为生理盐水组、氟尿嘧啶组、枸杞多糖高、中、低剂量组以及枸杞多糖加氟尿嘧啶组。实验结束后,检测各干预组脾指数、瘤重、肿瘤抑制率,MTT法检测淋巴细胞增殖反应、LDH检测NK细胞毒作用及中性红实验评估巨噬细胞吞噬能力,Griess法检测NO浓度,ELISA法测定TNF-α水平。结果枸杞多糖抑制荷S180小鼠肿瘤生长,刺激脾细胞增殖反应,提高NK细胞细胞毒效应以及巨噬细胞吞噬能力,刺激巨噬细胞NO分泌量,提高血清TNF-α水平。与氟尿嘧啶联合应用降低了氟尿嘧啶导致的免疫系统损伤。结论枸杞多糖可能是化疗药物的有效辅助药物。

Abstract:Objective To observe the effect of lycium chinensis mill polysaccharide(LBP)alone and in combination with 5-fluorouracil(5-FU) on the cytoimmunity and explore its immune regulatory mechanism in S180 mice.Methods Mice were randomly divided into a physiological saline group,a 5-FU group,a polysaccharide high,midddle and low dose group,and a polysaccharide plus 5-FU group.After the experiment,macrophage phagocytosis was detected by Neutral Red experiment and lymphproliferation response was detected by methyl thiazolyl tetrazolium(MTT) method.Cytotoxicity of NK cells to tumor cells was measured by LDH.No production was detected by Griess and TNF-α in the peripheral blood was inspected by ELISA.Results LBP alone inhibited S180 tumor growth,and its combination with 5-FU showed a synergistic effect.LBP stimulated the proliferation of lymphocytes,increased the cytotoxicity of natural killer cells,enhanced the phagocytosis and nitric oxide production,and increased the level of tumor necrosis factor TNF-α in the serum.In combination with 5-FU,LBP mitigated damage to the immune system caused by 5-FU in S180-bearing mice.Conclusion LBP might be a potential adjuvant for chemotherapy drugs.
