托吡酯片在人体内的药物动力学及生物等效性研究

目的研究托吡酯片的药物动力学及生物等效性。方法按两制剂双周期自身对照交叉试验设计,18名男性健康受试者分别单剂量口服托吡酯片参比制剂和受试制剂,用HPLC-MS/MS法测定血药浓度,计算药物动力学参数,并评价两制剂的生物等效性。结果口服托吡酯片参比制剂和受试制剂100 mg后的主要药物动力学参数：Cmax分别为（1 507.7±381.9）和（1 483.3±480.1）ng mL-1;tmax分别为（2.00±1.23）和（2.00±1.29）h;AUC0～t分别为（51 681±15 264）和（49 599±13 286）ng h mL-1;AUC0～∞分别为（57 198±16 599）和（55 345±14 423）ng h mL-1;t1/2分别为（35.57±6.12）和（36.63±7.98）h;托吡酯的相对生物利用度F0～t为（96.9±10.9）%,F0～∞为（97.8±11.4）%。结论 2种托吡酯片制剂具有生物等效性。

Pharmacokinetics and bioequivalence of topiramate tablets in healthy volunteers

Objective To determine the pharmacokinetics and bioequivalence of topiramate tablets.Methods A single oral dose of 100 mg reference and test topiramate tablets was given to 18 healthy volunteers in a randomized 2-way cross-over study.The pharmacokinetics as well as relative bioavailability were measured.Results The main pharmacokinetics of the reference and test tablets were as follows： Cmax（1 507.7±381.9） and（1 483.3±480.1） ng mL-1;tmax（2.00±1.23） and（2.00 ±1.29） h;AUC0 ～ t（51 681±15 264） and（49 599±13 286） ng h mL-1;AUC0 ～∞（57 198±16 599） and（55 345 ±14 423） ng h mL-1;t1/2（35.57±6.12） and（36.63±7.98） h.The relative bioavailability of F0 ～ t was（96.9±10.9）% and F0 ～ ∞ was（97.8±11.4）%.Conclusion The reference and test topiramate tablets are bioequivalent.
