Rho/Rho激酶在COPD大鼠气道的表达及其对气道重塑的影响

摘    要:
目的探讨Rho/Rho激酶信号通路在慢性阻塞性肺疾病大鼠气道中的表达及其对气道的影响。方法清洁级相同重量级Wistar大鼠60只,随机分为正常对照组、COPD组、Y-27632干预组,每组20只。分析各组大鼠的支气管壁厚度(War)和平滑肌厚度(Warn),以及RhoA,ROCKlImRNA的表达。结果与对照组相比,COPD组大鼠RhoA,ROCK1ImRNA表达明显增高,wat、wam也明显增加(P〈0.05);与C()PD组相比,Y-27632干预组大鼠RhoA,ROCKIImRNA的表达明显降低,Wat、wam也明显降低(P〈0.05)。结论COPD组大鼠RhoA,ROCKⅡmRNA表达明显增高;应用ROCKⅡ特异性拈抗剂Y一27632能够明显控制RhoA,ROCKⅡmRNA的表达,从而抑制慢性阻塞性肺疾病大鼠的气管壁增厚和平滑肌增殖。

Abstract:
Objective To investigate the Rho / Rho pathway expression in rat airways in chronic obstructive pulmonary disease and its impact on the airway. Methods 60 of clean-grade Wistar rats with the same body weight were randomly divided into normal control group,COPD group and Y 27632 the intervention group(n=20). Analysis of the bronchial wall thickness of the rats in each group(Wat)and smooth muscle thickness(Wam), RhoA, ROCK Ⅱ mRNA expression. Results Compared with the control group, the RhoA and ROCK Ⅱ mRNA in expression was significantly higher in the COPD group rats. At the same tirne,Wat and Warn also significantly increased(P〈0.05) ; compared to COPD group,of Y 27632 intervention group the role of RhoA and ROCK II mRNA, significantly lower expression,and the Wat,Wam were significantly lower than in COPD group rats(P〈0.05). Conclusion COPD rats have higher RhoA, ROCK Ⅱ mRNA expressions; application ROCK Ⅱ specific antagonist Y-27632 significantly control the expression of RhoA and ROCK Ⅱ mRNA, thus inhibiting the Wat and Wam of chronic obstructive pulmonary disease in rats.
