摘    要:目的研究不同浓度的大豆异黄酮对体外培养大鼠成骨细胞增殖活性和骨钙素(BGP)基因表达的影响。方法采用新生大鼠颅骨分离培养成骨细胞,加入不同浓度的大豆异黄酮(20、40、60、80μg/μL),另设空白对照组。MTT法测成骨细胞增殖活性。提取细胞总RNA,RT-PCR半定量分析细胞BGP mRNA表达。结果 MTT结果显示,60和80μg/μL组成骨细胞增殖活性与对照组相比显示增高,有统计学意义(P<0.05)。RT-PCR结果显示:60和80μg/μL组BGP mRNA表达增强,有统计学意义(P<0.01)。结论大豆异黄酮可以促进成骨细胞的增殖活性和BGP基因上调,并呈明显的剂量依赖关系。

Abstract:Objective To study the effect of different concentrations of isoflavones on gene expression of BGP mRNA and cell proliferation activity in cultured rat osteoblasts. Methods Newborn rat calvarid osteoblastic cell were isolated and cultured. They were cultured in medium with various concentration isoflavones(20, 40, 60, 80 μg/μL), compared to control group. The proliferation activity by MTT method. The total RNA was extracted by TRIZOL and analyaed with semi-quantitative RT-PCR. Results Compared to control group, the cell proliferation activity of 60 and 80 μg/μLgroup was significantly promoted(P〈0.05), the cell BGP mRNA expression of 60 and 80 μg/μL group was significantly upregulated(P〈0.01). Conclusion The isofiavones could increase cell proliferation activity and expression of BGP mRNA. The effect was similar to estrogen.
