摘    要:目的探讨Wnt-2、β-catenin和TCF-4在非小细胞肺癌组织中的表达特点及其与肺癌临床组织分型、分化程度以及与淋巴结转移的相关性。方法应用免疫组化SP方法检测45例非小细胞肺癌组织、10例癌旁正常组织中Wnt-2、β-catenin和TCF-4的表达情况;应用RT-PCR及Western blot技术分别检测这三个指标在mRNA和蛋白水平的表达情况。结果 (1)Wnt-2、β-catenin和TCF-4在非小细胞肺癌组织中表达阳性率分别为64.4%、75.6%和80%,β-catenin和TCF-4在低分化和中高分化间存在差异(P<0.05),β-catenin在淋巴结转移组间存在统计学差异(P<0.05);(2)定位于Wnt信号通路中的三个关键因子Wnt-2、β-catenin和TCF-4之间表达具有相关性。结论Wnt-2、β-catenin和TCF-4在非小细胞肺癌中异常高表达,提示其代表的Wnt信号通路活化。

Abstract:Objective To investigate the expression of Wnt-2,β-catenin and TCF-4 in non-small cell lung cancer and evaluate their relationship with histological type,differentiation cell and lymphoid node metastasis.Methods Immunohistochemical staining with SP method was conducted to identify the expression rates of Wnt-2,β-catenin and TCF-4 in the lung tissue near tumor(5 cm beside the tumor) in 10 cases and in the non-small cell lung cancer tissue in 45 ones.By the methods of Western Blotting and RT-PCR,a paralleled study was conducted to check the expression or distribution of Wnt-2,β-catenin and TCF-4 in the above tissues.Results(1) The positive expression rates of Wnt-2,β-catenin and TCF-4 were 64.4 %,75.6% and 80.0%,respectively.The expression of β-catenin and TCF4 were both related with differentiation(P< 0.05),and that of β-catenin was related with tumor diameter.(2) There were positive correlations among the expression rates of Wnt-2,β-catenin and TCF-4 in non-small cell lung cancer(P< 0.05).Conclusion Abnormal expression of Wnt-2,β-catenin and TCF-4 in non-small cell lung cancer tissues indicates that the Wnt signal pathway is activated.
