| 摘 要： | 目的对KM小鼠胃Cajal间质细胞进行体外分离、培养,并对其进行鉴定,摸索高效实用的方法,为进一步探索研究Cajal间质细胞的移植、替代治疗及药物实验提供基础.方法以KM株小鼠胃为组织来源,体外分离、培养ICC,并进行细胞传代,光镜下观察其形态,用荧光标记C-Kit特异性抗体标记细胞,进行免疫组化鉴定.结果细胞稳定贴壁后,形成梭形或三角形突起,3 d后可出现长突起,并可逐渐形成网络连接.结论酶解法分离细胞成功,但纯度低,数量不足;体外培养ICC 24 h后逐渐稳定贴壁,3 d以后可形成网络连接,可稳定培养3周以上;传代后细胞正常形态逐渐发生变化,20 d后可逐渐凋亡.
