白藜芦醇对刀豆蛋白A所致肝损伤的保护作用

摘    要:目的探讨白藜芦醇对刀豆蛋白A(ConA)所致肝损伤的保护作用。方法用ConA诱导小鼠肝损伤,用白藜芦醇灌胃治疗,3d后取血,检测血清ALT、AST活性和一氧化氮(NO)含量,检测肝组织超氧化物歧化酶(SOD)的活性和丙二醛(MDA)含量,并观察肝组织病理学变化。结果白藜芦醇中剂量组ALT、AST活性均明显低于模型组(P<0.01),但与联苯双酯组比较,差异无显著性意义(P>0.05);白藜芦醇大、中剂量组肝组织SOD活性明显升高、MDA含量显著降低,与模型组比较,差异有显著性意义(P<0.05);白藜芦醇大、中、小剂量组血清NO含量较模型组明显降低(P<0.05);白藜芦醇中剂量组肝组织病理改变明显减轻。结论白藜芦醇对ConA所致小鼠肝损伤具有较好的保护作用,其保肝机制与抗脂质过氧化损伤和抑制NO的产生密切相关。

Abstract:Objective To investigate the protective effects of resveratro l on liver injury in mice induced by concanavalin A(Con A). Methods The model o f liver injury induced by Con A was used for evaluating the therapeutic effects of resveratrol in mice. After 3 days of resveratrol was given to mice by gastric perfusion, their blood samples and liver tissues were collected. The serum tran saminase(ALT,AST) activity and nitric oxide(NO) level were determined. The path ological changes of liver was observed. The super oxide dismutase(SOD) activity and malondialdehyde(MDA) level in liver tissue were also determined. Results Se rum ALT activity, AST activity and NO level in middle dosage group were obviousl y lower than that in model group(P<0.01), but no significant difference comparin g with that in bifendate group(P>0.05).It showed that SOD activity and MDA conte nt in liver tissue significant decreased in both high and middle dosage groups ( P<0.05). NO content in all the three groups of different dosage of resveratrol w as obviously lower than the model group (P<0.05). Hepatic histopathological chan ges were remarkable alleviated in middle dosage group. Conclusion Resveratrol h as a good protective effect on liver injury in mice induced by Con A. The protec tive effect may be due to antilipoperoxidation and suppressing the release of NO .
