实验性2型糖尿病大鼠病证结合模型的建立

目的:从中西医结合角度,探讨实验性2型糖尿病大鼠病证结合模型的建立。方法:Wistar大鼠70只随机分为空白组30只和模型组40只,空白组给予基础饲料并腹腔注射等剂量柠檬酸缓冲液,模型组给予高热量、高糖、高脂饮食饲养并一次性腹腔注射链脲佐菌素(STZ)至12周。观察两组大鼠体重和外观行为变化。结果:模型组大鼠第4周开始体重呈明显持续增加,第12周下降,但仍明显高于正常组(P<0.05);外观行为学变化:模型组大鼠第4～8周被毛脱落、倦怠、懒动、嗜睡,饮食量1～8周增加,9～12周减少,饮水量增加,大便干结,臀尾出汗潮湿;第11～12周出现臀毛枯黄,尾淤斑,舌紫暗。结论:高热量、高糖、高脂饮食配合STZ腹腔注射建立实验性2型糖尿病大鼠模型,随造模时间延长表现出由气阴两虚逐渐发展为气阴两虚兼痰浊(血淤)证演变趋势,具有中医证候动态变化特点。

Establishment of Rat Models Coupled with Disease and Symptom of Type 2 Diabetes

Objective:From the standpoint of combination of TCM and western medicine,we discussed the establishment of rat models coupled with disease and symptom of type 2 diabetes.Methods:The 70 Wistar rats were randomly divided into the blank group(30 rats)and the model group(40 rats).The blank group were fed with basal feed and given peritoneal injection citric acid buffer at the same dosage,the model group were fed with high heat,high glucose and higher fatty acid with peritoneal injection of STZ twelve weeks.The changes in appearance,behavior and weight of rats were observed.Results:The weight of the model group gained up obviously at 4th week,and lowers continually from the 12th week,higher obviously than the blank group.Appearance and behavior:during the 4th week to 8th week,the model rats appeared in loss of hair,accidie,lethargy,diet increased but decreased during the 9th week to 12th week,drinking increased,dry stool,sweatiness and wateriness at tail.During the 11th week to 12th week,the model rats appeared loss of hair and turn into yellow color,dark-purple tongue,and some petechias appeared at the tail from top to root.Conclusion:The rat model of type 2 diabetes established by feeding high heat,high glucose and higher fatty acid with peritoneal injection of STZ,appeared the evolution trend of syndromes in Chinese medicine is from the simultaneous deficiency of both qi and yin to both qi and yin with turbid phlegm(blood stasis).It had the characters of dynamic evolutionary in traditional Chinese medicine syndromes.
