Ⅰ型糖尿病患者的HLA-DR基因单体型分析

目的：揭示中国人ＩＤＤＭ的遗传分子基础。方法：应用分子生物学研究方法，对ＨＬＡ－ＤＲＢ进行序列特异性引物的ＰＣＲ扩增，鉴定了８１例ＩＤＤＭ患者和８４例正常对照汉族人群的ＤＲＢ基因多态性。结果：ＩＤＤＭ患者中ＤＲＢ１＊０３，ＤＲＢ１＊９等位基因频率明显高于对照组，其频率分别为８．６４％Ｖ．Ｓ３．０％和２８．４％Ｖ．Ｓ１６．１％（Ｐ〈０．０５）；ＩＤＤＭ患者中ＤＲＢ１－ＤＲＢ３／ＤＲＢ１－ＤＲＢ４基因

Analysis of HLA-DR Haploid Genes in IDDM patients

Objection:To indicate the genetic molecular basis in Chinese.Method: We Applied molecular biological technique to do the specula amplification on HLA-DRB using PCR. and identified the Dan gene polymorphism in 81 patients with IDDM and that of 84 normal control from Han Nationality. Results: The frequency. of DRB1*03,DRB1*09 in patients with IDDM was obviously higher than that of controls. The frequency was 8.64%V.S 3.0% and 28.4% V.S 16. 1% respectively (P<0.05),The frequency of DRB1- DRB3/DRB1-DRB4 in patients was obviously higher than of controls, 19.8% V.S 6.7%(P<0.05), We identified the that there was positive correlation between DRB1*12 allele and IDDM in Chinese.Conclusion: DRB1*03 and DRB1*09 are the susceptible genes Chinese; DRB1 * 12 is the inferior susceptible gene; DRB1* 15 and DRB1*16are the resist genes.
