强力霉素对制动兔骨关节炎模型对侧膝关节早期退变的影响

摘    要:目的:研究口服强力霉素对制动兔骨关节炎模型对侧膝关节早期退变的影响及作用机制。方法:36只新西兰大白兔随机分为空白对照组,模型对照4、8、12周组,给药8、12周组。模型组与给药组均给予石膏固定左下肢。石膏固定4周后,给药组给予强力霉素。分别在8周、12周处死动物,观察指标包括:软骨大体形态、软骨man—kin评分、软骨Ⅱ型胶原、uPA、MMP-13。结果:与模型对照组比较,给药组软骨面光滑,mankin评分较低,Ⅱ型胶原含量较高,uPA、MMP-13活性较低(P〈0.01)。结论:强力霉素对制动兔骨关节炎模型对侧膝关节软骨早期退变有缓解作用,作用机制与抑制MMP-13、uPA的表达有关。

Abstract:Objective: study on The effect and action mechanism of Doxycycline on the early degeneration of opposite knee of the braking rabbit model osteoarthritis. Methods:Totally 36 New Zealand rabbits were assigned into 6 groups:blank control group, Model 4 week group, Model 8 week group, Model 12 week group, dministration 8 week group, administration 12 week group. Both Model group and administration group were given left lower extremity fixed by gypsum. After 4week,administration group were given Doexyeyclin. Six animals were sacrificed in 8 and 12 weeks after fixation. The observed indexes include the degeneration of articular cartilage , Mankin score, the expression of Collagen Type Ⅱ , the expression of Collagen,uPA,MMP- 13. Results:compare with the model group,the cartilage of administration group is smooth,Mankin score Low, high content Collagen Type Ⅱ, low activities of uPA, MMPs - 13 ( P 〈 0.01 ). Conclusion: Mitigative Effect of Doxycycline on the early degeneration of opposite knee of the Immobilizied rabbit model osteoarthritis, The action mechanism was positively related with the expression of uPA and MMPs - 13.
