摘 要: | <正> Peary等将LPS(革兰氏阴性细菌脂多糖)与小鼠脾细胞在体外一同培养时,发现培养基中DNA合成显著增强,从而证实了LPS对小鼠脾细胞的致有丝分裂作用。进一步的实验证实LPS仅为B淋巴细胞的致有丝分裂原。其作用的确切机理,仍未完全明了。目前为人们接受的理论是,LPS的这一作用是通过B淋巴细胞膜上的致有丝分裂受体来实现的。

In Vivo Transformation of Murine Spleen Cells Stimulated by Bacterial IPS |

Abstract: | In vivo and vitro transformation of marine spleen lymphocytes stimulated by LPS was compared. In vitro test was carried out by adding 100 ng of LPS to 5 × 106 cells in 1 ml RPMI 1640 medium. After 24-hour incubation, the incorporation of 3H-TdR started and reached its peak at 72 hours, and then decreased rapidly.The in vivo test was done by IP injection o?lOOUg of LPS in each mouse and a group of mice were killed at an interval of 24 hours. The spleen cells were incubated with 3H-TdR for 20 hours and the radioactivity was counted. The incorporation of 8H-TdR into cells started at 48 hours after LPS injection and reached its peak at 96 hours, and then declined slowly.The in vivo transformation test was based on the knowledge that the message of LPS stimulation murine spleen cells receive in vivo can be expressed through 3H-TdR uptake by murine spleen cells in vitro. This method is useful for the study of kinetics of lymphocyte transformation stimulated by LPS in vivo. |
