| 摘 要： | 目的探讨铜对肝癌细胞株HepG2的IL-8分泌及其mRAN表达的影响。方法分别用ELISA法和RT-PCR测定不用铜添加组HepG2细胞上清液中IL-8浓度和细胞内IL-8mRNA表达丰度。结果在一定范围内培养细胞上清中IL-8水平和细胞内mRNA表达丰度与培养基中铜浓度成正比,而铜离子超过一定浓度后二者水平反而降低。结论在一定范围内铜元素可使肝癌细胞的IL-8分泌和表达增加,铜对肝癌的影响可能是通过促使血管生成因子IL-8的分泌与表达。

| Abstract: | Objective To study the effect of copper ion on hepatocarcinoma cell HepG2 IL-8 mRNA expression and IL-8 release. Method ELISA and RT-PCR were used to determine the IL-8 and mRNA expression of HepG2 cell with different copper ion concentrations. Results Under certain concentration range of copper ion, the level of IL-8 and mRNA expression were in direct proportion with the copper ion levels, however, when copper exceeded certain level, their expression and release were inhibited. Conclusion Under certain concentration range, copper ion stimulates IL-8 release and mRNA expression, which might be the possible mechanism that copper ion effects on hepatocellular carcinoma (HCC). |
