聚乙二醇修饰超氧化物歧化酶的基础研究

摘    要:目的研究聚乙二醇修饰的超氧化物歧化酶基本特性的改变。方法以聚乙二醇为修饰剂,对猪红细胞超氧化物歧化酶进行共价修饰。比较超氧化物歧化酶修饰前后在免疫原性,血浆存留时间,抗酶水解能力等方面的改变。结果聚乙二醇的修饰,使超氧化物歧化酶的免疫原性消失,体内作用时间延长,抗酶水解能力增强。结论聚乙二醇修饰的超氧化物歧化酶稳定性较高。

Abstract:Objective To study the effect of polyethylene glycol(PEG) of 5000 daltons was attached covalently to available lysine residues of pig erythrocyte superoxide dismutase.Methods Using cyanuric chloride as the coupling agent.The modified superoxide dismutase preserved of the activity of native enzyme.Results Its plasma half-life increased from 15 mim to about 30 h in the mouse during intravenous injection.Modified superoxide dismutase appeared to lose its antigenicity,it did not react with antibodies raised against native superoxide dismutase.Conclusion After modified SOD with polyethylene glycol was stable.
