血管内皮细胞生长因子促进猪真皮下血管网皮片成活的实验研究

摘    要:目的 研究重组人类血管内皮细胞生长因子(vascular endothelial growth factor,VEGF_(165))对真皮下血管网皮片(free skin grafts with preserved subcutaneous vascular network,PSVN)成活的作用。方法 在8只纯白色家猪脊腹两侧各形成6个真皮下血管网皮片,分别厚为3、4和5mm。分别在对照组皮片下和实验组皮片下局部应用生理盐水和VEGF_(165)。术后12d,测量各组PSVN皮片的成活率及皮片新生血管密度。结果 术后12d时,3mm实验组皮片成活率及新生血管密度与对照组相比无显著性差异(P>0.05);4mm和5mm实验组皮片的成活率为76.15%和50.08%,明显优于对照组的35.61%和13.49%(P<0.05)。4mm和5mm实验组皮片真皮下新生血管密度为14.34个/mm~2和4.30个/mm~2,明显优于对照组的8.60个/mm~2和0.72个/mm~2(P<0.05)。结论 人类重组VEGF_(165)能够提高带有较厚皮下脂肪的真皮下血管网皮片的成活。

Abstract:Objective To investigate the applied value of recombinant human VEGF_(165) to improve the viability of free skin grafts withpreserved subcutaneous vascular network. Methods Eight white pigs were used, on back each pig had 6 PSVN graft, whose thickness was 3,4 and 5 mm respectively, and 48 PSVN grafts were divided randomly into 2 groups: the control group received 0.9% NaCl 3 mL;the treatmentgroup received VEGF_(165) 120 ng locally. On postoperative 12 day, the survival area of each graft was measured and grafts were harvested forhistological analysis, and the density of angiogenesis was also examined. Results On postoperative 12 day, the mean skin survival rate of 4mm grafts and 5 mm grafts were 76.15% and 50.08% in experimental group; 35.61% and 13.49% in control group. The density ofangiogenesis of 4 mm grafts and 5 mm grafts were 14.34/mm~2 and 4.30/mm~2 in treatment; 8.60/mm~2 and 0.72/mm~2 in control. Comparedwith control group. the mean survival rate and the density of angiogenesis of the group treated with VEGF_(165) was of statistical significance (P<0.05). Conclusion The results suggested recombinant human VEGF_(165) improve the viability of free skin grafts with preserved thicksubcutaneous vascular network.
