醋酸铅对体外培养人肾小球系膜细胞超微结构的影响

摘    要:
目的探讨醋酸铅对人肾小球系膜细胞(human renal mesangial cells,HRMC)超微结构的影响。方法醋酸铅处理HRMC后,Giemsa染色法观察细胞形态学变化;透射电镜观察细胞超微结构的变化;DNA梯(DNA Ladder)实验检测醋酸铅对HRMC基因组的影响;流式细胞仪检测细胞凋亡率。结果 10μmol/L醋酸铅染毒HRMC 24、48h后,Giemsa染色显示出细胞成凋亡形态学改变。透射电镜观察细胞胞质内出现空泡,核固缩,核膜结构模糊,并出现凋亡小体。HRMC基因出现较为明显的DNA损伤现象。流式细胞仪检测细胞凋亡率较对照组明显升高(P<0.05)。结论醋酸铅可促进HRMC凋亡,影响肾脏正常功能。

Abstract:
Objective To explore the toxic effect of lead acetate on ultrastructure of human renal mesangial cells(HRMC).Methods After HRMCs were treated with lead acetate,Giemsa staining was applied to observe the morphological changes of these cells.Transmission electron microscope was used to observe the ultrastructure changes.DNA damage of HRMC was detected by DNA ladder test.The rate of apoptosis was measured by the flow cytometry.Results After treated with 10 μmol/L lead acetate for 24 h,and 48 h,characteristic changes of apoptosis in HRMC were observed in the treatment group.Under electron microscopy,vacuoles in the cytoplasm and karyopycnosis could be observed,and the nuclear membrane was vague,and an increased number of apoptotic bodies were found.DNA ladder test showed diffused bands appeared in the treatment groups compared with the control group.The flow cytometry results showed higher cell apoptosis rate induced by the treatment group than that of the control group(P<0.05).Conclusions Lead acetate could remarkably inhibit human renal mesangial cells growth and promote apoptosis affecting the kidney function.
