Zebularine对Jurkat、K562细胞p53基因表达的影响

摘    要:目的了解药物zebularine对白血病细胞株Jurkat、K562细胞p53基因表达的影响,探讨其作用机制及在白血病基因治疗中的价值。方法根据zebularine作用终浓度将白血病细胞株Jurkat、K562各分为3组:对照组、250μmol/L组、500μmol/L组,分别用RT-PCR、Western-blot方法检测Jurkat、K562细胞在药物干预48 h后p53 mRNA和蛋白的表达情况。根据检测结果,探讨其作用机制及应用价值。结果在zebularine干预48 h后,在对照组和250μmol/L组均检测不到p53mRNA表达,而在500μmol/L组中检测到有p53mRNA表达,差异有显著性(P<0.05);6组细胞样品中均检测到p53蛋白表达,在500μmol/L组中表达上升,差异有显著性(P<0.05)。结论Zebularine可促进Jurkat、K562白血病细胞株p53的表达,并有一定的浓度依赖性,提示zebula-rine的干预激活了p53的表达,为zebularine治疗白血病提供了实验室依据。

Abstract:Objective To investigate the mechanism and value of zebularine in gene therapy for leukemia by studying the influence of zebularine on p53 expression in leukemia cell lines Jurkat and K562.Methods The leukemia cell line Jurkat and K562 were divided into three groups respectively: control group(zebularine was not added),250 μmol/L group(the final concentration of zebularine was 250 μmol/L) and 500 μmol/L group(the final concentration of zebularine was 500 μmol/L).The expression of p53 mRNA and protein after 48 h zebularine intervention were measured by RT-PCR and Western blot,respectively.Results The expressions of p53 mRNA were not detected in both of the control groups and 250 μmol/L groups in Jurkat and K562,but were detected in the 500 μmol/L groups.The differences were statistically significant(P<0.05).The expression of p53 protein were increased in 500 μmol/L groups,and the differences were statistically significant(P<0.05).Conclusions The expression of p53 in leukemia cell lines Jurkat and K562 can be promoted by zebularine.The effect is partially concentration-dependent.These results provided the experimental basis of zebularine treatment for leukemia.
