马桑果致小鼠肝脏损伤初步研究

摘    要:目的研究马桑果对健康昆明小鼠肝脏的损伤作用,为探讨马桑果的毒性以及对机体的损伤机制提供依据。方法选取健康昆明种小鼠80只,随机分为空白对照组、5、10、20 g/kg BW染毒组,每组20只,雌雄各半。建立小鼠急性染毒模型,空白对照组用蒸馏水灌胃,记录灌胃后临床表现,在试验后24 h处死小鼠,采集血清,检测天门冬氨基转移酶(aspartate aminotransferase,AST)、丙氨酶氨基转移酶(alanine aminotransferase,ALT)、碱性磷酸酶(alkaline phosphatase,ALP);解剖小鼠取肝脏,计算肝脏的脏器系数,HE染色观察各组小鼠的脏器组织病理学改变。结果24 h处死的小鼠中,20 g/kg BW染毒组肝脏脏器系数明显高于空白对照组及5、10 g/kg BW染毒组,差异均有统计学意义(均P<0.05);20 g/kg BW剂量组雄性小鼠的ALT、AST、ALP 3个值与空白对照组及5、10 g/kg BW剂量组相比较有明显升高趋势,差异均有统计学意义(均P<0.05)。20 g/kg BW剂量组的雌性小鼠ALT、ALP的值显著高于空白对照组、5 g/kg BW剂量组,差异均有统计学意义(均P<0.05)。20 g/kg BW剂量组的AST值显著高于空白对照组及5、10 g/kg BW,差异有统计学意义(P<0.05)。10和20 g/kg BW染毒组中多数小鼠肝脏出现明显损伤,呈现病变程度随着剂量增加而加重的趋势,肝脏损伤表现为肝脏充血,细胞水肿,脂肪病变,炎症细胞浸润。在空白对照组未发现异常病变。结论马桑果对小鼠肝脏有明显的损害作用,提示除了神经系统,肝脏也是马桑果中毒的靶器官之一。

Abstract:Objective To investigate the toxic effect of coriaria sinica fruit on the liver of healthy kunming mice,and to provide clues for the study of the toxicity of coriaria sinica fruit and the mechanism of injury to the body.Methods A total of 80 healthy kunming mice were randomly and equally divided into blank control group,5,10 and 20 g/kg BW poisoning group,10 mice in each group.The blank control group was irrigated with distilled water.The clinical manifestations after perfusion were recorded.Mice were killed 24 hours after the test and serum was taken.Aspartate aminotransferase(AST),Alanine aminotransferase(ALT),and Alkaline phosphatase(ALP)were tested.The organ coefficients of the liver was calculated.The histopathological changes liver was observed by HE staining.Results The liver organ coefficient of the 20 g/kg BW poisoning group was significantly lower than that of the blank control group and the 5,10 g/kg BW poisoning group(P<0.05);the ALT,AST,and ALP values of male mice in the 20 g/kBW dose group were higher than those in the blank control group,5 g/kg BW,and 10 g/kg BW groups(P<0.05).The value of ALT and ALP in female mice in the 20 g/kg BW dose group was higher than that in the blank control group and the 5 g/kg BW group(P<0.05).The AST value of female mice in the 20 g/kg BW dose group was higher than that of the blank control group,5 g/kg BW,and 10 g/kg BW(P<0.05).Some mice in the 10 and 20 g/kg BW group showed obvious liver damage,and the degree of lesion increased with the dose.Liver injury mainly includes liver congestion,cell edema,fatty lesions,inflammatory cell infiltration.No abnormal lesions were found in mice of negative control group.Conclusion Coriaria sinica can cause obvious damage to the liver of mice,suggesting that besides nervous system,the liver is also one of the target organs of Coriaria sinica poisoning.
