血栓通对脑缺血再灌注损伤大鼠神经细胞HSP70表达的影响

摘    要:目的观察血栓通对大鼠脑缺血再灌注损伤神经细胞热休克蛋白70(heat shockprotein 70,HSP70)表达的影响,探讨其神经保护作用机制。方法将大鼠随机分为5组,其中川芎嗪组术前3 d腹腔注射川芎嗪注射液80 mg/kg,血栓通大、小剂量组术前3 d分别腹腔注射血栓通10 mg/kg和5 mg/kg。除假手术组外,其余4组用双侧颈总动脉结扎法建立大鼠脑缺血再灌注损伤模型,于脑缺血30 min再灌注24 h后断头取脑,用免疫组织化学法检测脑组织中HSP70表达,并与病理组织学改变进行对照。结果血栓通能明显增强大脑皮质及海马HSP70的表达,减轻神经细胞损伤。结论血栓通能减轻脑组织的缺血再灌注损伤,改善神经功能缺失,其作用机制可能与增加HSP70表达有关。

Abstract:Objective It is to observe the influence of Xueshuantong on nerve cell heat shock protein 70(HSP70) expression in rats with cerebral ischemia reperfusion injury and to study its nerve protection mechanism.Methods The rats were randomly divided into five groups.Ligustrazine group was given Ligustrazine injection 80 mg/kg by intraperitoneal injection three days before operation,and Xueshuantong large dosage and small dosage groups were given Xueshuantong 10 mg/kg and 5 mg/kg respectively by the same method above.The rats of all the groups except notho-operation group were made into cerebral ischemia reperfusion injury models by bilateral common carotid artery ligation method.All the rats were killed and their brains were taken out after 24 h reperfusion injury.The HSP70 expression in brain tissue was measured with immunohistochemical method and was compared with the change of pathohistology.Results Xueshuantong could obviously increase the HSP70 expression in cerebral cortex and hippocampus and relieve nerve cell injury.Conclusion Xueshuantong can protect brain tissue from cerebral ischemia reperfusion injury and improve the nerve function.The mechanism may be related with the increasing of HSP70 expression.
