| 摘 要： | 目的：研究获得人体核迁移蛋白C在毕赤酵母中的高效表达的方法。方法：将重组表达质粒pPICZαA-hNUDC-his用BxtXⅠ单酶切,电击转入Pichia pastoris KM71,采用Zeocin筛选获得转化子。结果：通过优化诱导表达条件使目的蛋白获得较高的表达量。结论：通过优化诱导蛋白表达条件可大幅提高易降解蛋白hNUDC在毕赤酵母KM71中的表达量。 |

| Abstract: | Objective：To explore a method for efficient expression of obtaining human nuclear transfer protein C in the pichia.Methods：Recombinant expression plasmid pPICZα A-hNUDC-his was restricted by single enzyme BxtXⅠ,was shifted Pichia pastoris KM71 by electric shock and obtained transformants by adopting Zeocin screen.Results：There was higher expression of purpose protein by optimizing induced expression conditions.Conclusion：Higher expression of easily degradable protein hNUDC can be enhanced substantially in Pichia pastoris KM71 by optimizing induced expression conditions. |
