Rg1、Rb1对糖尿病肾病大鼠肾结缔组织生长因子的影响

目的:探讨Rg1、Rb1对糖尿病肾病(DN)大鼠肾结缔组织生长因子(CTGF)蛋白表达的影响。方法:雄性SD大鼠75只,按体质量随机分为正常组10只,模型组65只。模型组大鼠按55 mg/kg体质量腹腔一次性注射链脲佐菌素,72 h后血糖≥16.7 mmol/L,尿糖≥4"+"者为糖尿病造模成功。将造模成功的大鼠按血糖高低随机分为模型组、厄贝沙坦组、Rg1大剂量组、Rg1小剂量组和Rb1组。用药12周,处死大鼠无菌取肾,行肾组织HE、Mallory染色,观察各生化指标和肾组织病理形态学改变,采用免疫组化方法分析肾组织CTGF蛋白的表达情况。结果:模型组肾组织表现为DN病理改变;正常组及用药组肾小管上皮细胞、肾间质细胞胞浆有轻度CTGF表达,模型组CTGF则呈强阳性表达,Rg1、Rb1组CTGF表达较模型组明显减少(P<0.05)。结论:Rg1、Rb1可通过对肾组织CTGF蛋白表达下调而发挥治疗和预防糖尿病肾病的作用。

Effects of Ginsenoside-Rg1,Rb1 On connective tissue growth factor of diabetic nephropathy rats

Objective:To observe effects of Rg1,Rb1 on protein expressions of connective tissue growth factor(CTGF) of diabetic nephropathy rats.Methods:75 SD rats were randomly divided into normal control group(n=10)and model group(n=65).Model group rats by 55 mg/kg one-time were intraperitoneal injected streptozotocin,72 h after glucose≥16.7 mmol/L,urinary glucose≥++++,for diabetes modeling success.The modeling success rats were redivided into model group,irbesartan group and Rg1 group and Rb1 group.Administration for 12 weeks,the rats were sacrificed,observed pathological changes of renal tissue,using immunohistochemistry analysis renal tissue CTGF protein expression.Results:Model group showed renal tissue DN pathological changes,normal group and the treatment group tubular epithelial cells of renal and renal interstitial cells had light CTGF expression,there was a strong expression in model group.Rg1 group and Rb1 group and the model group CTGF expression more than the model was significantly reduced(P<0.05).Conclusion:Rg1,Rb1 play the role of treatment and prevention DN by down-regulating protein expression of CTGF.
