灵芪胶囊对S180荷瘤小鼠p53和bcl-2蛋白表达影响的实验研究

[目的] 探讨灵芪胶囊对S180荷瘤小鼠的抑瘤作用及其抑瘤作用机制.[方法]采用体内实验法,于无菌条件下抽取传代7 d、生长良好的S180即荷瘤小鼠腹水,无菌生理盐水1:3稀释(约含瘤细胞2×109/L,),按0.2 mL/只接种于消毒后的小鼠右前肢腋部皮下,制成肿瘤模型,经口灌胃灵芪胶囊11 d后,称取小鼠体质量,脱椎处死小鼠,取脾脏及胸腺称质量并计算胸腺指数和脾脏指数.用免疫组化法检测各组凋亡相关基因突变型p53和bcl-2基因的表达.[结果]灵芪胶囊在明显抑制肿瘤生长的同时不降低体质量,在-定剂量对荷瘤动物的免疫器官指数无明显影响,明显优于化疗药物;灵蓖胶囊3个剂量组突变型p53基因和bcl-2基因的表达均小于模型组.[结论]灵芪胶囊对S180荷瘤小鼠有明显抑瘤作用,其机制可能与抑制突变型p53基因和bcl-2基因表达有关.实验证明灵芪胶囊不失为-种很有前途的抗癌中药.

Effect of Lingqi-capsule on gene expression of p53 and bcl-2 on sarcoma S180-bearing mice

[Objective] To discuss the antitumor effect of Lingqi-capsule on sarcoma S₁₈₀-bearing mice.[Methods] In vivo experiment wasAdopted and ascites sample from sarcoma S₁₈₀-bearing mice grew for 7 days in asepsis condition.The sample was diluted for 1:3 withasepsis brine (about included sarcoma cell 2×10⁹/mL), inoculated into skin of right forelegs with 0.2ml per mice for establishing themodel of sarcoma S₁₈₀-bearing mice.After giving Lingqi-capsule for 11 days the immunity organs index and the gene expression of p53 and bcl-2 in each group were compared with the model group.[Results] Lingqi-capsule could obviously inhibit tumor growing withoutlosing of body weight and influencing immune organs.The gene expression of p53 and bcl-2 in each group was less than that of the modelgroup.[Conclusion] Lingqi-capsule can obviously inhibit sarcoma S₁₈₀ mice growing.The mechanism of action may be correlated withdecreasing of p53 and bcl-2 gene expression.The aim of this thesis was looking for a new medicine having effect both raising immunityand antitumor from Chinese herbal medicine.Lingqi-capsule was proved to be a promising medicine by this experiment.
