非小细胞肺癌COX-2蛋白表达及与bcl-2相关性

摘    要:
 目的 探讨非小细胞肺癌 (NSCLC)环氧化酶 2 (cyclooxygenase 2 ,COX 2 )蛋白表达的临床意义及与bcl 2的关系。方法 免疫组织化学法检测 5 3例NSCLC癌组织及配对癌旁正常组织COX 2蛋白的表达及定位 ,Western印迹法半定量检测COX 2蛋白表达水平 ;免疫组织化学法检测癌组织bcl 2蛋白表达。结果 肺癌组织COX 2蛋白表达高于癌旁正常组织 (P <0 .0 1) ,腺癌高于鳞癌 (P <0 .0 5 ) ,但与患者的年龄、性别、TNM分期及肿瘤组织的分化程度无明显相关 (P >0 .0 5 )。COX 2与bcl 2蛋白表达相关 (P <0 .0 5 )。结论 COX 2在肺癌组织尤其是肺腺癌中表达上调 ,COX 2与bcl 2在NSCLC发展中可能起协同作用。

Abstract:
Objective To investigate the expression of cyclooxygenase 2 (COX-2) protein and its relationship with bcl-2 in non-small cell lung cancer(NSCLC). Methods Immunohistochemical assay was used to detect the localization of COX-2 protein expression in cancerous tissues and paired noncancerous tissues from 53 patients, and the semi-quantitative COX-2 protein was evaluated by Western blot. bcl-2 protein was detected by immunohistochemical assay. Results Expression of COX-2 protein was significantly higher in cancerous tissues than paired noncancerous tissues( P < 0.01 ). Expression of COX-2 protein was higher in adenocarcinoma than in squamous carcinoma ( P < 0.05 ). There was no significant relationship between COX-2 protein expression and patients'age, sex, TNM stages and tumor histological differentiation ( P > 0.05 ). The expression of COX-2 protein was correlated with bcl-2 protein expression( P < 0.05 ). Conclusion COX-2 was up-regulated in lung cancer tissues especially in adenocarcinoma, COX-2 and bcl-2 may play a synergetic role in the process of NSCLC carcinogenesis.
