促黄体素释放激素对大白鼠睾丸的组织学和组织化学观察

用81只雄性大白鼠分为9组,分别给予LHRH-A_3 0.02μg、2μg 及200μg 三种剂量注射2天,20天及60天。在组织学上见200μg 注射20天后,睾丸开始出现抑制退化现象。200μg 注射60天后,则抑制现象更加明显。睾丸重量下降,曲细精管萎缩,生精细胞疏松脱落,精子细胞溶解,精母细胞膨大变形,形成严重生精障碍。在组织化学上,注射LHRH-A_3 200μg 60天后,可见睾丸的新陈代谢受到影响,物质的吸收及转运出现抑制,生精上皮有一定损伤,睾酮的合成增强但分泌受到障碍。提示注射LHRH-A_3 200μg 60天后,对大鼠精子生成和睾酮的分泌发生明显的抑制作用。

HISTOLOGICAL AND HISTOCHEMICAL STUDIES OF THE TESTES IN RATS TREATED WITH LUTEINIZING HORMONERELEASING HORMONE

Nine groups of 9 male rats each were injected with 0.02μg,2μg and 200μg of lutein-izing hormone releasing hormone(LHRH-A_3)daily for 2,20 and 60 days.The groupinjected with 200μg of LHRH-A_3 for 20 days exhibited initial degenerative changes in theseminiferous epithelium,while for 60 days the inhibitory effects became more pronounced.There were decrease in the weight of testis,atrophy of smeiniferous tubules,loosening andexfoliation of seminiferous epithelium,degeneration and dissolution of spermatids,swellingand deformation of spermatocytes and serious hindrance of spermatogenesis.After 200μgof LHRH-A_3 for 60 days,SDH and GPDH activities indicated histochemically thatLHRH-A_3 interfered with the metabolism of testes.The decrease of NSE and AIP activityshowed that LHRH-A_3 affected the secretion of testosterone and absorption and transportof materials.The increase of ACP activity was related to the removal of injured cells in thetestis.The enhancement of 3β-HSDH activity indicated increase in testicular biosynthesisand inhibition of gonadotrophin secretion.All of the above suggest inhibition of spermato-gensis and secretion of testosterone following administration of 200μg of LHRH-A_3for 60 days.
