摘    要:用本室建立的染色体原位切口移位方法,详细描述了人淋巴细胞染色体上DNase-1敏感区的分布。与已知的G带、R带图谱和六对已发表的染色体D带图谱比较,表明所揭示的B带与G带、R带和D带有明显的差异,染色体原位切口移位后的DNase—1敏感区多分布在具有转录活性的常染色质区,而异染色质区则通常与失活的基因部位有关。作者对B带的生物学意义和应用价值进行了详细讨论。

Abstract:Mapping of B-bands on the in situ nick-translated human chromosomes completely substituted with 5-bromodeoxyuridine has been presented in comparison with the G-, R-and D-bands.It is demonstrated that the light B-bands are usually related to euchromatin region of chromosomes, while the dark B-bands to heterochromatin region. The B-bands are not only reverse D-bands, but also significantly different from G- and R-bands in banding patterns on the human chromosomes.In accordance with the analysis of localization of DNase-1 sensitive regions on the human metaphase chromosomes, the biological significances of B-band mapping in molecular biology and molecular genetics may be revealed as : 1) understanding the gene expression in eukaryotic cells in the chromosomal level, 2) elucidating the molecular mechanisms of some human malignancies and genetic disorders, 3) reflecting the active state of genes in different mammalian genomes, 4) exploring the relationship of gene activity with cell type, cell differential degrees, cell growth and developement.
