顺铂诱导大鼠卵巢早衰的实验研究

目的腹腔注射不同剂量的顺铂制备卵巢早衰(POF)大鼠动物模型,摸索成模的最佳致病剂量。方法成熟雌性Wistar大鼠分5组腹腔注射不同剂量的顺铂,A组3 mg/kg×7 d,B组2.5 mg/kg×7 d,C组2.0 mg/kg×7 d,D组2.5 mg/kg×5 d,E组予生理盐水,建立大鼠卵巢早衰模型。观察大鼠体质量、动情周期、血清性激素水平、脏器系数及形态学改变。结果腹腔注射顺铂后,A、B、C、D、E组大鼠病死率分别为70%,50%,30%,0,0。顺铂组体质量均明显下降,大鼠动情周期延长。C组血清FSH水平显著高于E组(P<0.05),D组与E组无显著性差异(P>0.05)。血清E2水平C、D 2组均显著低于E组(P<0.05)。卵巢及子宫指数C组显著低于E组(P<0.05),肾上腺指数C组显著高于E组(P<0.05)。顺铂组大鼠卵巢原始卵泡数和初级卵泡数均明显降低,而闭锁卵泡数均明显增加。结论 2.0 mg/(kg.7 d)模型组是最理想的卵巢早衰动物模型。此化疗损伤性卵巢早衰大鼠模型血FSH明显升高,卵巢组织学衰退性改变与人类化疗损伤性卵巢早衰病变过程相似。

Experimental study of cisplatin inducement in establishment of the rat model of premature ovarian failure

Objective It is to explore the optimal dosage of cisplatin inducement in establishment of the rats model of premature ovarian failure(POF).Methods Adult female Wistar rats were divided into 5 groups by treated with the different doses of cisplatin to establish models of POF:group A 3 mg/kg for 7 days,group B 2.5 mg/kg for 7 days),group C 2.0 mg/kg for 7 days),group D 2.5 mg/kg for 5 days) and with saline in group E.The changes of general state,weight,genesial hormones,organs index and pathological changes were observed in the rats after the injection of cisplatin.Results The weight as higher and the duration of estrous cycle as longer in all cisplatin groups after injection than before the injection.In groupA,B,C,D,E the mortalities were 70%,50%,30%,0,0.For the serum FSH levels,there was no statistical significance between group E and group D(P>0.05),but significantly higher in group C(P<0.05).The serum E2 levels in groups C and D were significantly lower than that in group E(P<0.05).Index of ovarian and uterus in group C was significantly lower than that of group E(P<0.05),but adrenal index in in groups C was significantly higher.The numbers of original and primary follicles were significantly reduced in cisplatin groups,but the numbers of atretic follicles were significantly increased.Conclusion The incretion and changes of biochemial indexes are similar to those in clinic in mice injected 2.0 mg/(kg·7 d)of CDDP.This method of model establishment is most suitable for systematic study on POF.
