摘    要:树突状细胞属于白细胞系,其功能为捕捉抗原并将该抗原呈递给T淋巴细胞而引发免疫应答。本研究在包装细胞中克隆了一个突变的erbB-2基因,并将该基因导入到CD34~ 来源的树突状细胞中。经检测,该基因失去了致癌性,不能转化NIH3T3细胞,但能表达较高水平的突变蛋白,组化染色和流式细胞术都确证了这一点。经基因修饰的树突状细胞将在乳腺癌的免疫治疗中具有潜在的应用价值。

Abstract:The dendritic cells (DC) belong to the lineage of white blood cells, which specialize in capturing antigens and stimulating T lymphocytes-dependent immunity. Genetically modified DCs present tumor antigen to T lymphocytes to elicit killing of tumor cells, so that DC can be considered as " natural vaccine". In order to develop a protocol to kill residual breast cancer cells in body after bone marrow transplantation by strengthening immune surveillance of T lymphocytes, a mutant erbB-2 gene was cloned and transduced this mutant gene into human CD34 cell-derived DC using spinoculation method coated with fibronectin material to facilitate retrovirus gene transter efficiency. It was verified that transduced DC expressed relatively high level of the mutant erbB-2 protein, which was eradicated of the ability to transform mouse NIH3T3 cells. This gene-modified DC possess potential to stimulate T lymphocytes for immunotherapy in the future.
