摘    要:目的调查不同类型自体吞噬在肝癌及癌旁组织中的分布情况。方法通过电子显微镜观察15对肝癌组织标本的超微结构,并使用相关软件量化图像指标以分析不同类型自体吞噬分布与细胞超微结构特征之间的联系。结果数据提示肝癌细胞中主要分布巨型自噬,分布与肿瘤细胞的活动及代谢情况相关;而癌旁细胞主要分布微型自噬,且分布与细胞的炎症损伤程度相关。结论巨型自体吞噬和微型自体吞噬在肝细胞癌组织及癌旁组织中呈现不对称分布,该现象可能与肝癌的发生相关。

Abstract:Aims: To investigate the distribution of various type of autophayg in liver tumor and nontumor tissue, and analyze the complicated role of of autophagy in HCC. Methods: We observed ultrastructure of 15 pairs of HCC tumor and matched nontumor tissue by EM, quantitated the morphometric data with soft of Scion Image, and analysed the relative between distribution of various types of autophagy and ultrastructure feature of 158 tissue cells. Categorical data were analyzed using SAS software.Results: The data showed asymmetric distribution of macroautophagy and microautophagy between liver tumor cells and hepatocytes, the activity of microautophagy was frequent in hepatocytes and absent in tumor cells, the distribution of macroautophagy was just opposite. We also found the correlation between activity of microautophagy and status of cells inflammation. And the activity of macroautophagy might be correlated with stress of energy.Conclutions: Our study suggests that the asymmetric distribution of various autophagy contributes to the dual roles of autopahgy; As housekeeping function of cells, the macroautophagy maintains viability in liver tumor cells during periods of metabolic stress. On the other hand, microautophagy maintains homeostasis in hepatocytes to suppress tumorigenesis.
