| 摘 要： | 目的探讨含菊藻丸大鼠血清对人激素非依赖性前列腺癌细胞系PC-3增殖和细胞周期的影响。方法以3种不同质量浓度的菊藻丸水剂对成年雄性去势SD大鼠灌胃。另以等量生理盐水灌胃进行对照。末次灌胃后于60～90min时间段内取下腔静脉血制成血清,用以体外培养PC-3;用MTT法测定培养4、24、48、72h的细胞存活率,用流式细胞仪测定培养48h的细胞周期分布。结果高、中剂量的含药血清可抑制PC-3细胞的生长,且抑制作用随时间的延长而增强,低剂量无明显抑制作用。含药血清高剂量组G1-G0期细胞比例低于中、低剂量组,G2/M期细胞比例高于中、低剂量组。结论含菊藻丸血清能抑制PC-3细胞增殖,可用于动物实验进一步明确体内有无抗肿瘤作用;不同质量浓度含药血清对细胞周期影响不同,机制有待研究。
