摘    要:
目的:人脐带间充质干细胞用PKH26标记,探讨人脐带间充质干细胞在大鼠化疗性卵巢早衰模型中的卵巢迁移示踪。方法:提取并培养人脐带间充质干细胞,按PKH26标记程序进行细胞标记,透射电镜观察标记组和未标记组细胞的超微结构,测定细胞增殖、周期、凋亡情况。荧光显微镜下观察PKH26标记的人脐带间充质干细胞移植后的卵巢情况。结果:标记后细胞的形态无明显差别,均为成纤维细胞样,细胞增殖未见明显影响,细胞生长状态良好,PKH26标记阳性的细胞主要存在卵巢血管中。结论:PKH标记技术可用于人脐带间充质干细胞移植治疗化疗所致卵巢早衰中的归巢、分化、增殖的研究。

Abstract:
Objective:To track PKH26-labeled human umbilical cord mesenchymal stem cells(MSCs)after transplanting into rats of chemotherapy-induced premature ovarian failure.Methods:Isolated human umbilical cord mesenchymal stem cells were labeled with PKH26,and cell ultrastructure was observed under transmission electron microscope.Cell cycle,cell apoptosis,expression of PKH26 and the proliferation rate were evaluated.The migration of PKH26-labeled human umbilical cord mesenchymal stem cells in ovary of the rat model was observed under fluorescence microscope.Results:There were no detectable differences in cell morphology,cell growth and proliferation after PKH26 labeling as human umbilical cord mesenchymal stem cells exhibited fibroblast-like spindle-shape.PKH26-1abeled cells were mainly distributed in blood vessel of stroma ovarii.Conclusion:PKH26 labeling technology may be useful for studying homing,differentiation,and proliferation of human umbilical cord mesenchymal stem cells after transplantation into rats of chemotherapy-induced premature ovarian failure.
