摘    要:目的探讨模拟失重对人胃黏膜上皮HFE-145细胞形态、增殖、周期和凋亡的影响.方法采用回转器模拟失重,用图像分析系统软件分析HE染色后人胃黏膜上皮HFE-145细胞形态的改变,用免疫组化法观察增殖细胞核抗原(PCNA)表达的变化,用流式细胞仪测定细胞周期,并用TUNEL法检测细胞凋亡.结果回转组HFE-145细胞在模拟失重12 h时的面积较对照组增大(P<0.01),其余时间段细胞的面积与对照组无明显差异;其长短径之比在各时间段与对照组均无明显差异.与正常对照组相比,回转组HFE-145细胞24 h、36 h、48 h及72 h的PCNA抗体阳性细胞的比率无明显差异,12 h回转组PCNA抗体阳性细胞比率明显减少(P<0.05).24 h、36 h、48 h和72 h回转组HFE-145细胞的细胞周期分布与对照组相比无明显差异,12 h回转组G0+G1期细胞比例较对照组明显增多(P<0.05),S+G2+M期细胞之和较对照组显著减少(P<0.05).24 h、36h、48 h和72 h回转组HFE-145细胞凋亡的比例与对照组相比无明显差异,12 h回转组细胞凋亡比例较对照组明显增多(P<0.05).结论回转器模拟失重使胃黏膜上皮HFE-145细胞在12 h时形态发生了变化,出现细胞周期阻滞、增殖抑制和凋亡增加.

Abstract:Objective To investigate the effects of simulated weightlessness on cellular morphology, proliferation, cell cycle and apoptosis of human gastric mucosa cell line HFE-145. Methods Human gastric mucosa cell line HFE-145 were cultured in vitro and divided into 2 groups: simulated weightlessness group and control group. A rotating clinostat was used to simulate weightlessness. Specimens were examined after rotating for 12 h, 24 h, 36 h, 48 h and 72 h respectively, and compared with controls. Image-pro4.5 image analysis system was employed for morphometric study. The expressions of proliferating cell nuclear antigen (PCNA) were examined with immunohistochemical stain. The changes of cell cycle were examined by a cytometer. Apoptosis was measured by TUNEL method. Results Compared with control group, the area of cell line HFE-145 increased significantly after 12 h simulated weightlessness(P<0.01), but did not change significantly at other time points, the ratio of long diameter and short diameter did not change significantly at all time points. Compared with control, the expression of PCNA of cell line HFE-145 did not change significantly after 24 h, 36 h, 48 h and 72 h simulated weightlessness, but decreased significantly after rotating 12 h (P<0.05). Compared with control, the cell cycles of cell line HFE-145 after rotating 24 h, 36 h, 48 h and 72 h did not change significantly, but the cells in G_0 G_1 phase in 12 h group increased significantly (P<0.05), and the cells in S G_2 M phase decreased significantly (P<0.05). Compared with control, the apoptosis index of cell line HFE-145 in 24 h, 36 h, 48 h and 72 h groups did not change significantly, but increased significantly in 12 h group (P<0.05). Conclusions The cellular morphology of HFE-145 cell was changed after rotating in a clinostat for 12 h. The conversion of cell from G_1 phase to S phase was blocked, the proliferation was inhibited, and the apoptosis index was increased after rotating for 12 h.
