聚维酮联合多柔比星灌注对膀胱癌组织中Fas和Caspase-3 mRNA表达的影响

摘    要:目的探讨聚维酮(PVP)联合多柔比星膀胱灌注对膀胱肿瘤组织中凋亡相关蛋白Fas、Caspase-3mRNA表达的影响。方法选取术前24h膀胱灌注生理盐水30mL加多柔比星20mg(对照组),10%PVP30mL加20mg多柔比星(实验组),术后病理证实为浅表性膀胱肿瘤标本各25例。采用RT-PCR方法检测肿瘤组织中凋亡相关蛋白Fas、Caspase-3mRNA的表达水平。结果FasmRNA和Caspase-3 mRNA在对照组中的阳性率较实验组高,且差异存在高度统计学意义(P〈0.01)。结论PVP联合多柔比星膀胱灌注能上调凋亡相关蛋白Fas、Caspase-3mRNA的表达。

Abstract:A/M To observe the expressions of Fas mRNA and Caspase-3 mRNA after bladder instillation of polyvinylpyrrolidone combined with doxonlbicin. METHODS Patients with bladder cancer were divided into two groups randomly. They received intravesical instillation with doxorubicin 20 mg and NS 30 mL(control group) or doxorubicin 20 mg and 10% polyvinylpyrrolidone 30 mL (experimental group), respectively 24 h before operation. Twenty-five patients of each group were chosen after they were identified as superficial bladder cancer. The expressions of Fas mRNA and Caspase-3 mRNA of tumor tissue were determined by RT-PCR. RESULTS The expressions of Fas mRNA and Caspase- 3 were higher in experimental group than in control group. CONCLUSION Polyvinylpyrrolidone intravesical instillation combined with doxorubicin could up-regulate expression of Fas mRNA and Caspase-3 mRNA.
