银染—TRAP法检测胸腹水脱落细胞端粒酶活性的研究

目的:探讨检测胸腹水脱落细胞端粒酶活性对于鉴别良恶性肿瘤的临床意义.方法:用Kim法处理48例胸腹水细胞后,利用改进的银染—TRAP(端粒重复序列扩增)法测其端粒酶活性.结果:24例临床及细胞涂片检查证实恶性胸腹水细胞端粒酶活性全部阳性;12例临床证实恶性而细胞检查未找到癌细胞的胸腹水细胞端粒酶活性阳性7例,阴性5例;12例良性胸腹水细胞端粒酶活性全部阴性.结论:端粒酶的激活与恶性胸腹水的发生发展密切相关,并有可能成为一种鉴别良恶性胸腹水快速、灵敏的临床诊断指标.

Detection of telomerase activity in pleural and peritoneal effusion by silver staining telomeric repeat amplica-tion protocol

Objective:To explore the distinguish diagnosis of benign and malignant pleural and peritoneal effusion by detecting the telomerase activity. Methods: Combining silver stain with TRAP, we detected the telomerase activity of 48 exfoliated cells in pleural and peritoneal effusion. Results: Among 24 malignant exfoliated cells which were diagnosed by clinical and cell cytologicalexamination, telomerase activity was all positive (24/24). Among 12 malignant exfoliated cells which were only diagnosed by clinical examination, 7 were positive (7/12) . Telomerase activity was not detected in 12 negative controls. Conclusions: There is close relationship between Telomerase activity and malignant pleural and peritoneal effusion. Telomerase will be one of the marks to distinguish diagnosis of benign and malignant pleural and peritoneal effusion.
