复元胶囊对大鼠实验性骨关节炎软骨IL-6、bFGF的干预研究

目的:研究复元胶囊对大鼠实验性骨关节炎(OA)软骨白细胞介素-6(IL-6)和碱性成纤维细胞生长因子(bFGF)表达的影响。方法:Hulth法复制大鼠OA模型,随机分为对照、模型、四环素和复元胶囊高、中、低剂量组。灌胃给药,每天1次,连续10周。10周后取软骨标本,行大体观察、Mankin’s评分及免疫组化测定软骨中IL-6、bFGF的表达水平,并将IL-6、bFGF的表达水平与Mankin’s评分指数进行Pearson相关分析。结果:复元胶囊各组关节软骨Mankin’s评分、IL-6表达水平均低于模型组(P<0.01或P<0.05),bFGF表达水平均高于模型组(P<0.01);复元胶囊各剂量组Mankin’s评分差异具有显著性(P<0.05),IL-6表达水平与关节病理改变的Mankin’s评分指数呈正相关(P<0.01),bFGF表达水平与关节病理改变的Mankin’s评分无明显相关。结论:复元胶囊能下调大鼠OA软骨IL-6的表达,上调bFGF的表达,其作用呈剂量依赖性。高剂量复元胶囊抗大鼠OA作用与四环素相当。

Effects of Fuyuan Capsule on the Expression of Interleukin-6 and Basic Fibroblast Growth Factor in Rats with Experimental Osteoarthritis

OBJECTIVE： To observe the effects of Fuyuan capsule on the expression of interleukin-6 （IL-6） and basic fibroblast growth factor （bFGF） in rats with experimental osteoarthritis （OA）. METHODS： OA models were induced by Hulth technique. Model rats were randomly divided into six groups： control group, model group, tetracycline group, Fuyuan capsule high-dose, medium-dose and low-dose groups. Those groups were given relevant drug via i.g. gtt once a day for 10 weeks. Articular cartilages were obtained ten weeks later. Histomorphology of cartilage were evaluated according to macroscopic grading score and Mankin＇ s score was conducted. Expression of IL-6 and bFGF in articular cartilage were detected by immunochistochemistry technology. Pearson correlation analysis was carded out to determine Mankin＇ s index and the expression of IL-6 and bFGF in articular cartilage. RESULTS： Mankin＇s score and the expression of IL-6 in Fuyuan capsule groups were lower than in model group （P〈 0.01 or P〈0.05） ; the expression level of bFGF in Fuyuan capsule groups were higher than in model group （P〈0.01）; there was significant difference in Mankin＇ s score among Fuyuan capsule groups （P〈0.05）. Pearson correlation analysis showed that the expression of IL-6 in the articular cartilage was positively correlated with the pathological change of joint （P〈0.01）. There was no significant correlation between the expression of bFGF in the articular cartilage and Mankin＇s score. CONCLUSION： Fuyuan capsule can down-regulate the expression of IL-6 and up-regulate the expression of bFGF in dose-dependent manner. The treatment effect of high-dose Fuyuan capsule is identical to tetracycline in the treatment of OA.
