槲皮素对NB4,HL—60细胞形态、PML mRNA及蛋白表达的影响

摘    要:目的 探讨槲皮素对NB4,HL-60细胞的形态、PML mRNA及蛋白的表达影响。方法 以NB4,HL-60细胞为研究对象,细胞形态学观察采用Wright′s染色法及荧光染色法;PML蛋白细胞内分布定位采用免疫荧光技术;PML mRNA表达采用RT-PCR法。结果 1)经槲皮素处理后,各组细胞形态学上均出现凋亡特征性改变。2)槲皮素处理后NB4细胞内融合蛋白降解,PML聚积于POD结构,随后降解;在HL-60细胞,PML发生相似改变。3)槲皮素处理后各组细胞PML mRNA表达均无显著影响。结论 在槲皮素诱导下,PML在蛋白水平参与诱导白血病细胞凋亡机制;槲皮素能诱导,NB4,HL-60细胞凋亡,抑制白血病细胞生长。

Abstract:Objective To investigate the effects of quercetin on cell morphology, expression of PML mRNA and PML protein of NB4, HL 60 cells. Methods Cell morphology assayed by Wright's stain, fluorescence stain, and PML mRNA expression by RT PCR, PML protein localization by immunofluorescence.Results Typical apoptosis was found in NB4 and HL 60 cells after treatment with quercetin. Immunofluorescence analysis showed after treatment with quercetin, the fused protein degraded in NB4 cells, PML protein relocated, then degraded which was also seen in HL 60 cells. The expression of PML mRNA was not changed in quercetin treated cells.Conclusion PML plays a role as apoptosis inducer in leukemic cells at the translational level, quercetin can inhibit the proliferation of leukemic cells and induce apoptosis of NB4, HL 60 cells.
