Caveolin-1在骨肉瘤中的表达及其意义

目的观察Caveolin—1在骨肉瘤中的分布和表达情况，分析Caveolin-1在骨肉瘤临床诊断、治疗及预后评价中的意义。方法采用免疫组织化学SP法检测39例骨肉瘤组和22例良性骨肿瘤中Caveolin-1的表达，镜下观察Caveolin-1在肿瘤组织中的分布，应用自动图像分析检测Caveolin-1的表达情况。结果Caveolin-1表达于所有标本的肿瘤微血管和小血管内皮上，Caveolin-1表达于大多数骨肉瘤的肿瘤细胞，定位于细胞膜和胞浆，但在良性骨肿瘤中表达很微弱。Caveolin-1阳性表达率在骨肉瘤组中为59．0％，在良性骨肿瘤中为18．2％，两组间具有显著性差异（P〈0．01），有统计学意义，表明Caveolin-1在骨肉瘤中高表达。Caveolin-1的表达与骨肉瘤肿瘤体积、病理分型和患者的性别无关（P〉0．05），与骨肉瘤的组织分化和是否转移有关（P〈0．05）。结论Caveolin-1在骨肉瘤中高表达原因不明，其可能是促进骨肉瘤发生发展的一个潜在因素，也可能作为骨肉瘤治疗的新靶点。

The Expression and Clinical Significance of Caveolin-1 in Osteosarcoma

Objective To investigate the expression of Caveolin-1 in osteosarcoma and analyse its significance in diagnosis,treatment and prognosis of osteosarcoma. Methods Caveolin-1 was detected by SP immunohistochemical method in 39 cases of osteosarcoma and 22 cases of benign bone tumor,the distribution of Caveolin-1 in tumor tissues was observed under optical microscope,and the expression level of Caveolin-1 was detected by auto-image analysis. Results Caveolin-1 was strongly expressed in the endothelium of microvessels and small vessels in all cases,and mainly localized in the cell membranes and cytoplasm of the most osteosarcomatous tumor cells,Caveolin-1 was expressed weakly in benign bone tumor. The positive rate of Caveolin-1 in osteosareoma groups（59.0%） was significantly higher than that in benign bone tumor（18.2%）（P〈0.05）. The expression of Caveolin-1 was correlated with the metastasis of osteosareoma and tumor tissue differentiation, but not associated with the patients＇gender,tumor＇ size and the pathological type. Conclusion The reason why the most osteosareomatous tumor-cells strongly expressed Caveolin-1 was unknown ;Caveolin-1 is a potential factor during osteosarcoma growth and invasion ;Caveolin- 1 may be a new target for osteosareomatous therapy.
