基质金属蛋白酶2(9)在糖尿病患者动脉粥样硬化斑块中的表达及意义

摘    要:目的检测2型糖尿病患者动脉粥样硬化斑块内基质金属蛋白酶2(MMP2)和MMP9的表达和分布,探索MMP2和MMP9在糖尿病患者动脉粥样硬化斑块中表达的作用.方法应用免疫组织化学方法,分别检测MMP2和MMP9在13例健康人动脉,17例非糖尿病动脉粥样硬化患者及23例糖尿病患者动脉粥样硬化斑块内的分布和表达.结果抗MMP2、抗MMP9免疫沉积物主要集中在斑块核心周围,特别是在斑块的肩部和纤维帽.糖尿病组动脉粥样硬化斑块内抗MMP2免疫沉积物表达显著高于非糖尿病组(P<0.05);糖尿病组动脉粥样硬化斑块内MMP9表达也显著高于非糖尿病组(P<0.01).MMP2和MMP9在糖尿病患者动脉粥样硬化斑块中的表达明显高于非糖尿病患者.结论对动脉粥样硬化斑块不稳定性起重要作用的MMP2和MMP9,在糖尿病患者动脉中表达增强可能促进粥样硬化斑块的破裂.

Abstract:[Objective]To investigate the role of matrix metalloproteinases 2(MMP-2)and MMP-9 expression in atherosclerotic plaques of diabetic patients. The expression of MMP-2 and MMP-9 in the arteries from 23 diabetic patients, 17 non-diabetic patients with atherosclerosis and 13 normal controls was detected by immunohistochemical technique. Staining of MMP-2 and MMP-9 was mainly shown around the plaque core, especially in the plaque shoulder and thinned fibrous cap. Immunoreactivity for MMP-2 of diabetic patients with atherosclerosis was strikingly higher than that of the non-diabetic patients with atherosclerosis (P <0.05). Immunoreactivity for MMP-9 was significantly elevated in samples from diabetic group compared with samples from non-diabetic group with atherosclerosis (P <0.01). The expression of MMP2 and MMP9 in atherosclerotic plaques of diabetic patients is significantly higher than that of non-diabetic patients. [Conclusion] MMP-2 and MMP-9 accumulation,which play an important role in instabilizing plaques, may promote atherosclerotic plaque disruption in diabetic patients.
