新型免疫调节性寡聚脱氧核苷酸的序列设计和筛选分析

摘    要:
目的:探讨免疫调节性寡聚脱氧核苷酸(ODN)设计原则和筛选方法,为研究和开发新型免疫调节性ODN提供参考。方法:利用含有CpG基序的免疫刺激性寡聚脱氧核苷酸(CpG ODN)作为刺激物,将人外周血单个核细胞(hPBMC)作为研究对象,以3H-TdR掺入法检测CpG ODN诱导的细胞增殖水平,对自行设计的ODN的免疫调节活性进行筛选分析。结果:通过对两批免疫调节性ODN的设计和筛选,得到具有较强免疫抑制活性的ODN 667B,与CpG BW006组相比,其抑制率达到66.3%(P<0.01),并具有浓度相关性。结论:总结出免疫调节性ODN的设计原则,同时证明筛选方法可以用于免疫调节性ODN的筛选,为进一步研究开发新型免疫调节性ODN奠定了基础。
