大鼠肝细胞经脾脏多次移植的实验研究

目的　研究大鼠肝细胞经脾脏多次移植的有效性及其安全性。方法　观察同种异体移植组和同系移植组进行 3次经脾脏肝细胞移植后 ,移植肝细胞在受体组织内的存活量及形态学的变化。结果　在同系移植组 ,3次移植与 1次移植相比较 ,前者明显地增加了受体内存活的移植肝细胞量 (P <0 .0 1) ,未产生严重的并发症。同种异体移植组在环孢素 A作用下 ,与同系移植组相比较 ,两组移植效果无显著性差异 (P >0 .0 5 )。结论　大鼠肝细胞经脾脏多次移植可有效地增加受体内存活的移植肝细胞量 ;同种异体移植组在免疫抑制剂作用下和同系移植组相比 ,移植效果相似。

The experimental research of repeated intrasplenic hepatocyte transplantation

Objective To study the safety and efficacy of repeated intrasplenic hepatocyte transplantation. Methods Hepatocyte transplantations were performed 3 times at interval of 8 days.Cyclosporine A was administrated as immunosuprressor in allotransplantation group.The area of hepatocytes in spleen was analyzed by ImageQUA analyzer system on the photoes of H?E stained sections.Results Compared with single transplantation,multiple transplantations increased the transplanted hepatocytes mass in spleen significantly without serious complication( P < 0.01 ).Under immunosuprression by cyclosporine A no significant difference was founded between allotransplantation and syntransplantation groups.Conclusions Repeated hepatocyte transplantation can improve the quantity of transplanted hepatocytes significantly.With the immunosuprresion,allotransplanlation has almost the same efficacy as syntransplantation.
