摘    要:
目的:探讨褪黑素(MT)对大鼠脑损伤的保护作用及其机制。方法:采用自由落体撞击伤方法制作创伤性脑损伤模型,致伤后连续给予褪黑素,测定脑损伤后12h、24h、48h、72h大鼠脑组织谷胱甘肽过氧化物酶(GSH-PX)活性、丙二醛(MDA)和钙离子(Ca2+)含量并观察脑微细结构的变化。结果:创伤性脑损伤后脑组织GSH-PX活性降低,MDA及Ca2+含量升高,褪黑素能部分反转这种变化,脑水肿减轻。结论:褪黑素对创伤性脑损伤有保护作用,其机制可能与保护GSH-PX活性,减少脂质过氧化有关。

Protective effects and mechanisms of melatonin on brain contusion in rats

Abstract:
Objective:To investigate the protective effects and mechanisms of melatonin on brain contusion . Methods:72 Sprague-Dawley rats were divided into three groups. Traumatic braininjury has been developed through Feeney's method, melatonin was given after being injuried and mensurated the activities of GSH-PX and the contents of MDA and Ca^2+ in rat brain serum afterl2h, 24h,48h,and 72h of brain contusion. The morphological change was observed at the injured area by LM. Results: The activities of GSH-PX reduced in rat brain serum following traumatic brain injury, but contents of MDA and Ca^2+ increased. Melatonin treatment partly reversed these changes and reduced the brain edema. The effect of melatonin was also confirmed histopathologically. Conclusion:Melatonin is neuroprotective in traumatic brain injury, its probable mechanism is related to protect the activities of GSH-PX. Melatonin may be a valuable therapeutic agent in the treatment of brain contusion.
