利用组织芯片技术探讨免疫组化染色中二抗回收的可行性

目的应用组织芯片技术,在免疫组化超敏二步法中对二抗回收的可行性进行探讨。方法根据所测试抗体的不同,选取本院病理科48例中性甲醛固定、石蜡包埋的组织标本（包括正常及病变的组织）,用组织芯片制作仪构建1个组织芯片蜡块。用免疫组化超敏二步法试剂分别测试3种单克隆抗体（Vimentin、CK和Ki-67）。结果经平行的免疫组化染色后,在48例的试验中,未经使用的二抗与经一次使用后回收的二抗显色的结果相同或相似。结论在此次免疫组化染色测试中,组织芯片显示了其信息含量多、可比性强及省时、省力等优点,二抗显色的结果相同或相似（P〉0.05）,提示超敏二步法中二抗回收是可行的。

Using tissue microarrays technique to explore the feasibility of recovery of secondary antibody in immunohistochemical staining

Objective To explore the feasibility of recovery of secondary antibody in immunohitochemical staining by using tissue microarray technique.Methods According to different testing antibodies,48 neutral formaldehyde fixed and paraffin-embedded samples were chosen to test used secondary antibody in tissue microarray,each of which would be used to test the monoclonal antibody such as Vimentin,CK or Ki-67 by HRP detection system.Results The effect of staining with both unused secondary antibody and used secondary antibody recovered once were similar.Conclusion Secondary antibody which recovered from immunohistochemical staining is feasibility.
