T细胞死亡偶联基因8在大鼠局灶性脑缺血-再灌注损伤中的表达

目的观察大鼠脑缺血-再灌注后缺血半暗带内T细胞死亡偶联基因8(TDAG8)的表达。方法体重250～280g的SD成年雄性大鼠36只,随机均分为正常组(C组)、假手术组(S组)和脑缺血-再灌注组(IR组)。采用大鼠大脑中动脉内线栓阻断法(MCAO)模型。IR组大鼠缺血2h后再灌注24h。再灌注结束后对大鼠进行神经行为学评分并检测大鼠脑缺血半暗带内的TD-AG8mRNA及TDAG8蛋白表达。结果 IR组脑缺血半暗带内TDAG8mRNA及TDAG8蛋白表达明显高于C组和S组(P<0.01);C组与S组之间差异无统计学意义。结论 TDAG8可能参与了大鼠脑缺血-再灌注损伤过程。

Expression of T cell death-associated gene 8 in rats after ischemia-reperfusion

Objective To observe the expression of T cell death-associated gene 8 (TDAG8) in the penumbra of cortex in rats after focal cerebral ischemia reperfusion (IR).Methods Thirty-six male SD rats, weighting 250-280 g, were equally randomized into normal control group(group C), sham-operation (group S) and ischemia reperfusion group(group IR). The focal cerebral ischemia-reperfusion models were established by middle cerebral artery occlusion (MCAO) method inrats. Group IR was reperfused for 24 h after ischemia for 2 h. Neurological function was appraised and the levels of mRNA and protein expression of TDAG8 were detected at the end of reperfusion.Results Compared with groups C and S, the expression of TDAG8 mRNA and protein in the penumbra region increased significantly in group IR (P<0.01). There had no significant difference in the expression of TDAG8 mRNA and protein between group C and group S.Conclusion TDAG8 may be involved in the brain ischemia reperfusion process in rats.
