摘    要:
目的探讨粒细胞-巨噬细胞集落刺激因子(GM-CSF)对高氧引起的新生大鼠肺损伤的干预作用。方法 32只大鼠随机均分为四组:95%氧浓度+GM-CSF 9μg/kg(A)组、GM-CSF9μg/kg(B)组、95%氧浓度+生理盐水(C)组和生理盐水(D)组。7d后计算肺湿质量与干质量比(W/D),检测支气管肺泡灌洗液(BALF)中白细胞数量、单核细胞趋化蛋白1(MCP-1)及白细胞介素8(IL-8)水平,RT-PCR检测肺组织中MCP-1mRNA表达,HE染色进行肺损伤病理评分。结果与C、D组相比,A、B组肺损伤评分及W/D、BALF中白细胞和中性粒细胞计数、MCP-1及IL-8水平、MCP-1mRNA表达均明显增加(P<0.05);除MCP-1水平及其mRNA表达外,A组上述各指标与B组比较差异有统计学意义(P<0.05)。结论 GM-CSF对高氧暴露引起的新生大鼠肺损伤具有保护作用,这可能与损伤部位炎症反应受抑制有关。

Abstract:
Objective To investigate the effect of granulocyte macrophage colony-stimulating factor(GM-CSF) on the hyperoxia-exposed lung injury in neonatal rats.Methods Thirty-two rats were equally randomized into four groups of A(exposed with 95% oxygen plus GM-CSF 9 μg/kg intranasally injection),B(intranasally injected with GM-CSF 9 μg/kg),C(exposed with 95% oxygen plus normal saline intranasally injection) and D(normal saline intranasally injection).Seven days later,the wet/dry ratio(W/D) of lung tissues was calculated,and the number of leucocytes and the levels of monocyte chemoattractant protein-1(MCP-1) and interleukin-8(IL-8) in the bronchoalveolar lavage fluid(BALF) were detected.The expression of MCP-1 mRNA in the lung tissue and the pathological score of lung injury were measured by RT-PCR and HE staining,respectively.Results Compared with groups of C and D,the pathological scores of lung injury,the W/D,the numbers of leucocytes and neutrophils in the BALF,the levels of MCP-1 and IL-8 and the expression of MCP-1 mRNA were all obviously increased in groups of A and B(P<0.05).Except MCP-1 level and its mRNA expression,the indexes above were significantly different between group A and group B(P<0.05).ConclusionGM-CSF can protect the the lungs from hyperoxia-induced injury in neonatal rats,which may be related to the inhibition of inflammatory reaction.
