穹窿海马伞损伤对成年鼠学习记忆和海马GFAP阳性细胞的影响

摘    要:
目的:探讨穹窿海马伞(FF)损伤鼠学习记忆能力与海马胶质纤维性蛋白(GFAP)阳性细胞之间的关系。方法:切断SD成年大鼠左侧FF,用Y迷宫和免疫组化结合图像分析系统测试大鼠学习记忆能力和海马GFAP阳性细胞的变化情况及相互关系。结果:损伤2周后,损伤组损伤侧海马CA1区辐射层和齿状回分子层GFAP阳性细胞数密度(Nv)较正常组分别增多30.29%和30.15%(均P<0.01),胞体面积分别增加16.04%和19.42%(均P<0.01),齿状回分子层体密度(VcvB)增大19.40%(P<0.05)。经相关分析,大鼠学习记忆能力与海马CA1区GFAP阳性细胞Nv呈负相关(r=-0.836,P<0.01),与齿状回Nv呈负相关(r=-0.792,P<0.01)。结论:海马星形胶质细胞可能参与学习记忆过程。

Abstract:
ve:To investigate the relationship between the GFAP-positive neurons of hippocampus and the learning and memory in rats with Fimbria Fornix (FF) injury. Methods: The FF fiber tract in adult rats was unilaterally transected by a stereotactic knife cut. Y-maze procedure and immunohistochemistry method were used to quantitatively assess the change of and the relationship between the ability of learning and memory and GFAP-positive neurons of hippocampus. Results; 2 weeks after the FF fiber tract was transected, the Nv and the average area of astroglia in radiatum layer within the CA1 region of hippocampus increased by 30. 29% and 16. 04% respectively when comparaed with those of the control group. The Nv, VvCB and the average area of astroglia in the molecular layer of gyrus dentatus increased by 30. 15% , 19. 40% and 19. 42% respectively in comparison with those of the normal control. Correlation analysis showed that the ability of learning and memory in rat was negatively correlated with both the Nv of GFAP positive astrocytes in CA1 region ( r = -0. 836 ,P <0. 01 ) and the Nv in gyrus dentatus (r= - 0. 792 , P < 0. 01). Conclusion ; The results indicate that GFAP-positive neurons of hippocampus may involve in the learning and memory in rats.
