石斛合剂对糖尿病模型大鼠胰岛细胞凋亡及凋亡相关基因Bax、Bcl-2 mRNA表达的影响

摘    要:
研究石斛合剂对高糖、高脂+STZ造模糖尿病大鼠胰岛细胞凋亡及相关基因Bax、Bcl-2mRNA表达的影响。方法大鼠高脂、高糖饲养1个月后腹腔注射STZ(30mg)制备糖尿病模型,经石斛合剂(7.5、15.0、30.0g.kg-1)治疗2周后检测胰岛细胞凋亡及凋亡相关基因Bax、Bcl-2mRNA的表达。结果石斛合剂可显著地降低糖尿病大鼠空腹血糖(P<0.05或P<0.01);石斛合剂可显著减少糖尿病模型大鼠胰岛细胞凋亡(P<0.05),降低Bax mRNA表达(P<0.05),增加Bcl-2mRNA表达(P<0.05或P<0.01))。结论石斛合剂可降低血糖、减少大鼠胰岛细胞凋亡,可能与其降低糖尿病模型大鼠胰腺组织Bax、增加Bcl-2表达有关。更多还原

Abstract:
Objective To observe the effects of dendrobium compound on the apoptosis and mRNA expression of bax and bcl-2 in islet cells in diabetes rats.Methods The diabetes was induced by high fat and glucose feeding for 1 month,followed by intraperitoneal injection of streptozotocin(30 mg).The diabetes rats were treated with dendrobium compound(7.5、15.0、30.0 g·kg-1)for two weeks and the apoptosis and mRNA expression of bax and bcl-2 were detected in islet cells.Results Dendrobium compound significantly decreased fasting blood glucose concentrations,islet cell apoptosis and bax expression,but obviously increased bcl-2 expression(P<0.05 or P<0.01).Conclusion Dendrobium compound can reduce blood glucose concentrations and islet cell apoptosis through decreasing bax expression and increasing bcl-2 expression in pancreatic tissue in diabetes rats.
