复方灯盏头痛片提取物的活性筛选

摘    要:
[目的]对不同工艺制备的复方灯盏头痛片提取物进行药效学比较研究。[方法]采用小鼠水浴甩尾反应实验、醋酸致小鼠扭体实验、硝酸甘油致小鼠偏头痛模型以及利血平低5-HT化小鼠偏头痛模型,对不同提取工艺所得复方提取物进行药效学筛选。[结果]3种工艺提取物均能有效提高甩尾潜伏期,抑制小鼠的扭体反应,具有明显的镇痛作用,其中以醇提物组效果最为显著;在硝酸甘油致小鼠偏头痛实验中,醇提物高低剂量组均能明显减少小鼠挠头次数,水提物及醇水双提物只有高剂量组可明显减少小鼠挠头次数(P<0.05);利血平低5-HT化小鼠偏头痛模型实验中,3种提取物均可提高脑组织及血液中5-HT的含量,其中以醇提物提升效果最为显著。[结论]相同剂量的复方醇提物药效活性优于水提物及醇水双提物,醇提工艺为其较优提取工艺,可作进一步研究和开发。

Abstract:
[Objective] To compare pharmacological properties of different extracts of compound erigeron headache tablet and to select the optimum extracting technique. [Methods] The method of acetic acid twisting and thrilling test were adopted in the research of analgesic effect, one mice migraine model was induced by nitroglycerin(10 mg/kg) and the other was duplicated by subcutaneous injection of reserpine, and then were treated with the extracts of compound. [Results] The extracts of compound erigeron headache tablet could significantly prolong the latency of thrilling, also could decrease the number of twisting, and had an obvious analgesic effect, the alcohol extracts in high dosage group were more effective than the others. The experiments of therapeutical effect showed that the alcohol extracts could both reduce the number of scratching. Water extracts and alcohol-water extracts only worked in high dosage group(P<0.05). The extracts could significantly increased 5-HT level, and the alcohol extracts group were more effective than the others. [Conclusion] The pharmacological effects of alcohol extracts are better than the other two in the same dose, the extraction method of alcohol is the optimum extraction technique. It is worth further researching and developing.
